ABSTRACT

Introdução: As lesões nervosas periféricas (LNP) podem resultar em distúrbios motores e sensoriais alterando a funcionalidade do membro afetado, porém pouco se conhece a respeito dos efeitos da fotobiomodulação (FBM) com diodo emissor de luz (LED). Objetivo: Analisar os efeitos do LED sobre a funcionalidade da marcha de ratos Wistar pós LNP. Metodologia: Ratos Wistar foram submetidos a LNP por esmagamento de ciático e analisados nos seguintes grupos experimentais: (1) Controle; (2) LNP; (3) LNP+ LED (780 nm, potência média 40 mW, exposição radiante, energia por ponto, 3,2 J sobre o nervo ciático (LEDn); (4) LNP+ LED em nervo e região do músculo envolvido (LEDnm) e (5) LNP+ LED apenas em região do músculo (LEDm). Após 7, 14, 21 e 28 dias foram realizadas as análises de marcha utilizando o Índice Funcional Ciático (IFC). Resultado: Após 7 dias, os grupos tratados com LED apresentaram uma melhora da marcha em relação ao grupo Lesão, sendo essa melhora mais pronunciada no grupo LEDn. Após 14 dias, os grupos LEDn e LEDnm apresentaram valores semelhantes ao grupo controle e após 21 e 28 dias o IFC não apresentou diferenças entre os grupos experimentais. Conclusão: O LED aumentou a funcionalidade da marcha avaliada pelo IFC após 1 e 2 semanas pós LNP, especialmente quando foi usado na região nervosa associada ou não à região muscular.


Introduction: Peripheral nerve injuries (PNI) can result in motor and sensory disturbances altering the functionality of the affected limb, however not much is known about the effects of photobiomodulation (PBM) with light emitting diode (LED). Objective: We aimed to analyze the effects of LED on the gait function of Wistar rats after PNI. Methodology: Wistar rats were submitted to PNI by sciatic crush and analyzed in the following experimental groups: (1) Control; (2) PNI; (3) PNI+ LED (780 nm, mean power 40 mW, radiant exposure, energy per spot, 3.2 J on the sciatic nerve) (LEDn); (4) LNP+ LED on nerve and involved muscle region (LEDnm) and (5) LNP+ LED only on muscle region (LEDm). After 7-, 14-, 21- and 28-days gait analyses were performed using the Sciatic Functional Index (SFI). Results: After 7 days, the groups treated with LED showed an improvement in gait compared to the PNI group, with this improvement being more pronounced in the LEDn group. After 14 days, the LEDn and LEDnm groups showed similar values to the control group and after 21 and 28 days the SFI did not show differences between the experimental groups. Conclusion: LED increased the gait functionality evaluated by SFI after 1 and 2 weeks post-PNI, especially when it was used in the nerve region associated or not with the muscle region.

ABSTRACT

SUMMARY OBJECTIVE: to identify, through an integrative review, national studies published over the last ten years highlighting products and therapies used in burns. METHODS: integrative research with studies published in the last ten years. Including clinical studies describing the use of the already established or innovative therapies in burns and the results obtained, published in national journals in the last ten years. Excluding articles published before 2007 and those that did not present results regarding the use of products in burns. RESULTS: ten articles that met the inclusion criteria were selected. Collagenase, 1% silver sulfadiazine, and porous cellulose membrane were some of the therapies cited. CONCLUSION: the casuistry was low; however, the good results obtained with porous cellulose membrane and silver nanocrystalline dressing are highlighted, since they were used in a larger number of patients in the studies evaluated.


RESUMO OBJETIVO: Identificar, por meio de revisão integrativa, estudos nacionais publicados nos últimos dez anos que destaquem produtos e terapêuticas utilizados nas queimaduras. MÉTODOS: Pesquisa integrativa com estudos publicados nos últimos dez anos. Incluídos os estudos clínicos que descreveram a utilização de terapias já consagradas ou inovadoras em queimaduras e os resultados obtidos e publicados em periódicos nacionais nos últimos dez anos. Excluídos os artigos publicados antes de 2007 e os que não apresentaram resultados quanto ao uso de produtos nas queimaduras. RESULTADOS: Selecionados dez artigos que atenderam aos critérios de inclusão, sendo colagenase, sulfadiazina de prata 1% e membrana celulósica porosa algumas das terapias descritas. CONCLUSÕES: A casuística foi baixa, porém, ressaltam-se os bons resultados obtidos com a membrana celulósica porosa e o curativo com prata nanocristalina, em virtude de terem sido utilizados em um maior número de pacientes nos estudos avaliados.

Summary Objective: The aim was to evaluate the effectiveness of the experimental synergists muscle ablation model to promote muscle hypertrophy, determine the period of greatest hypertrophy and its influence on muscle fiber types and determine differences in bilateral and unilateral removal to reduce the number of animals used in this model. Method: Following the application of the eligibility criteria for the mechanical overload of the plantar muscle in rats, nineteen papers were included in the review. Results: The results reveal a greatest hypertrophy occurring between days 12 and 15, and based on the findings, synergist muscle ablation is an efficient model for achieving rapid hypertrophy and the contralateral limb can be used as there was no difference between unilateral and bilateral surgery, which reduces the number of animals used in this model. Conclusion: This model differs from other overload models (exercise and training) regarding the characteristics involved in the hypertrophy process (acute) and result in a chronic muscle adaptation with selective regulation and modification of fast-twitch fibers in skeletal muscle. This is an efficient and rapid model for compensatory hypertrophy.


Resumo Objetivo: Avaliar a eficácia do modelo experimental de ablação dos sinergistas para promover a hipertrofia muscular, determinar o período de maior hipertrofia, sua influência sobre os tipos de fibras musculares e determinar diferenças na remoção unilateral ou bilateral para reduzir o número de animais utilizados nesse modelo. Método: Após a aplicação dos critérios de elegibilidade para sobrecarga mecânica do músculo plantar em ratos, 19 artigos foram incluídos na revisão. Resultados: Ocorre maior hipertrofia entre os dias 12 e 15, o que torna o modelo eficiente para alcançar a hipertrofia rapidamente. O membro contralateral também pode ser usado, pois não houve diferença entre a cirurgia unilateral e bilateral, o que reduz o número de animais usados no experimento. Conclusão: O modelo difere de outros modelos de sobrecarga (exercício e treinamento) em razão das características envolvidas no processo de sobrecarga imposta (aguda), resultando em uma adaptação crônica muscular com modificação de fibras de contração rápida do músculo esquelético. É um modelo rápido e eficiente para se estudar hipertrofia compensatória.

ABSTRACT

Objective: To determine the effective remnant power after the passage of low intensity laser of two different wavelengths (660 and 780 nm) through buccinator muscle in anatomical hemifaces. Methods: Five human hemifaces were dissected, and a laser was shone in the central region of the muscle with a receiver placed on the opposite side. The hemifaces were irradiated using the same dosimetric parameters (40 mW, 1 W/cm2, continuous mode, 0.04 cm2) and at the same point in every muscle; remnant power was measured for each irradiation. Results: The average remnant power after irradiation with 660 nm (149 ± 15 µW) light was significantly lower (p<0.0001) than with that of 780 nm (380 ± 40 µW). Conclusion: The LIL of 780 nm presented greater remnant power in comparison to the 660 nm laser after passage through the buccinator muscle, indicating the latter ́s greater penetration capacity.


Objetivo: Determinar a potência remanescente após a passagem do laser de baixa intensidade em dois comprimentos de onda (660 e 780 nm) através do músculo bucinador em peças anatômicas dissecadas. Métodos: Dissecaram-se cinco hemifaces humanas expondo o músculo bucinador. Aplicou-se o laser na região central de cada músculo, e o receptor, para aferir a potência remanescente, foi posicionado na superfície intraoral contrária. As hemifaces foram irradiadas nos mesmos parâmetros dosimétricos (40 mW, 1 W/cm2, modo contínuo, 0,04 cm2) e no mesmo ponto em cada músculo, sendo a potência remanescente aferida a cada repetição. Resultados: A potência média restante após as irradiações de 660 nm (149 ± 15 µW) foi menor (p <0,0001) que com as irradiações de 780 nm (380 ± 40 µW). Conclusão: O LBI de 780 nm apresentou maior potência remanescente que o LBI de 660 nm, após passagem pelo músculo bucinador, denotando sua maior capacidade de penetração.

ABSTRACT

Introdução: O ultrassom terapêutico (US) é muito utilizado na prática clínica, mas há poucos estudos sobre seu efeito na regeneração muscular. Objetivo: Avaliar os efeitos do US sobre a atividade mitocondrial e diferenciação de células musculares C2C12, quando aplicado concomitantemente à indução do processo de diferenciação. Métodos: As células musculares foram submetidas ao processo de diferenciação pela adição de meio de cultura DMEM, suplementado com 2% de soro de cavalo, e receberam simultaneamente tratamento com US (pulsado a 20%, 3 MHz, 0,2 e 0,5 W/cm², 5 minutos). A atividade mitocondrial foi avaliada após 24h, 48h e 96h pelo método MTT, e a diferenciação celular após um e três dias pela atividade de creatina quinase (CK). Resultados: Não houve alteração da atividade mitocondrial e de CK nos grupos que receberam tratamento com US nos diferentes períodos avaliados. Conclusão: O US, nos parâmetros avaliados, não foi capaz de alterar a atividade mitocondrial e a diferenciação de células musculares C2C12.


Introduction: The therapeutic ultrasound (US) has been widely used in clinical practice, but there are few studies on its effect on muscle regeneration. Objective: To evaluate the effects of the US on mitochondrial activity and differentiation of muscle cells C2C12 when applied concomitantly the induction of the differentiation process. Methods: Muscle cells were subjected to differentiation process by addition of DMEM culture medium supplemented with 2% horse serum and received concomitant treatment with US (pulsed at 20%, 3 MHz, 0.2 and 0.5 W/cm², 5 minutes). The mitochondrial activity was assessed after 24, 48 and 96 hours by MTT assay and cell differentiation after one and three days for the activity of creatine kinase (CK). Results: There was no change in mitochondrial activity and CK in the groups receiving US treatment in different periods. Conclusion: In the evaluated parameters, the US was not able to change mitochondrial activity and differentiation of muscle cells C2C12.

ABSTRACT

Objetivo verificar se existe relação entre a presença de disfunção temporomandibular (DTM) relacionada à variabilidade da dimensão vertical de oclusão em crianças e adolescentes na faixa etária de 7 a 12 anos. Métodos foram avaliadas 96 crianças e adolescentes do Instituto Rogacionista em São Paulo. A pesquisa diagnóstica da disfunção temporomandibular foi por meio do Índice de Helkimo e exame clínico posterior. Para a mensuração dos dados referentes à dimensão vertical foram empregadas as distâncias comissura labial - canto externo do olho e base do nariz – mento. A comparação dos valores médios das medidas antropométricas entre os gêneros e os grupos com e sem disfunção temporomandibular foi realizada empregando-se a análise de variância (ANOVA) complementada pelo teste least significance diference. Resultados a dimensão vertical de oclusão demonstrou medidas distintas nas crianças e adolescentes em todas as faixas etárias avaliadas, foram observadas alterações significantes dessa medida nas idades de 10 e 12 anos de ambos os gêneros. Conclusão pode -se concluir que na amostra estudada houve relação direta entre a presença  de DTM e  Dimensão vertical de oclusão (DVO), correlação positiva entre as medidas  comissura labial – canto do olho externo e Násio – Mento no sexo feminino e alterações significantes na dimensão vertical de oclusão nas idades de 10 e 12 anos para ambos os sexos. .


Purpose to establish the relationship between the presence of temporomandibular disorders and variability of vertical dimension of occlusion aged between 7 and 12 years. Methods it was analyzed 96 children and adolescents of Rogacionista Institute in Sao Paulo, Brazil. The diagnostic investigation of temporomandibular disorders was through Helkimo index and subsequent clinical examination. To measure the data on the vertical dimension were employed distances labial - corner of his eye and nose base - ment. To compare the mean values of anthropometric measurements between genders and the groups with and without TMD, we used analysis of variance (ANOVA) complemented by least significance diference test. Results the vertical dimension of occlusion showed distinct measures in children and adolescents in all age groups studied, significant changes were observed for this measure at ages 10 and 12 years of both genders. Conclusion we can conclude that in this sample there was a direct relationship between TMD and DVO, positive correlation between the measures lip commissure – corner of eye and nasium - mentum in females and significant changes in the vertical dimension of occlusion at the ages of 10 and 12 years for both sexes. .

ABSTRACT

Studies indicate that the anabolic nandrolone decanoate (Deca-Durabolin(r)) can modulate cell cycle regulation, but little is known about its effects on muscle cells. Anabolic steroids are used, especially by athletes, to improve muscle mass and performance in the practice of exercises. The aim of this study was to evaluate the effect of the anabolic Deca-Durabolin(r) on the proliferation of skeletal muscle precursor cells C2C12. Cells were grown in Dulbecco's Modified Eagle Medium (DMEM), being supplemented with 10% Fetal Bovine Serum (FBS) and subjected to differentiation by the addition of 2% horse serum. They were incubated with anabolic at concentrations of 5, 10, 25 and 50 µM. The groups that received no anabolic or vehicle served as controls. The viability (proliferation) was evaluated by the MTT method (3-[4,5-Dimethylthiazol-2yl]-2,5-diphenyltetrazolium bromide; Thiazolyl blue) after one, three and five days of incubation. Three independent experiments were performed in each of the mentioned conditions, and the results were submitted to statistical analysis with significance level of p≤0.05 (ANOVA/Dunnett). Results showed no difference in viability between muscle cells treated with anabolic and the control cultures in all parameters. In conclusion, nandrolone, at the used concentrations, was not able to alter the viability of muscle C2C12 satellite cells...


Se utilizan los anabolizantes, en particular por atletas con el objetivo de aumentar la masa muscular y mejoría del desempeño. Este estudio tuvo como objetivo evaluar el efecto del anabolizante Deca- Durabolin(r) sobre la viabilidad (proliferación) de las células satélites musculares C2C12 inducidas a la diferenciación, imitando el proceso de reparación tras una lesión. Las células fueron cultivadas en medio Eagle modificado por Dulbecco (DMEM) suplementado con 10% de suero fetal bovino (SFB) y sometidas a diferenciación mediante la adición de 2% de suero de caballo y, simultáneamente, incubadas con el anabolizante en las concentraciones de 5 , 10 , 25 y 50 µM. En los grupos que no recibieron el anabolizante, ni el vehículo sirvió como controle . La viabilidad (proliferación) se evaluó después de uno, tres y cinco días, utilizando el método de MTT (3 - [4,5 - dimetiltriazol - 2 - il ] -2,5 difeniltetrazolio) . Se realizaron tres experimentos independientes, en cada condición citada, y los resultados sometidos al análisis estadístico con nivel de significación de p≤0,05% (ANOVA/Dunnett). Los resultados permitieron verificar que no hubo diferencia en la viabilidad entre células musculares tratadas con anabolizante e inducidas a diferenciación y culturas de controles que sólo fueron inducidas a diferenciación en todos los parámetros evaluados. En conclusión, el anabolizante decanoato de nandrolona, en las concentraciones evaluadas, no fue capaz de alterar la viabilidad de células musculares C2C12 durante el proceso de diferenciación...


Os anabolizantes são utilizados, especialmente por atletas, com o intuito de aumento da massa muscular e melhora do desempenho. Este estudo objetivou avaliar o efeito do anabolizante Deca-Durabolin(r) sobre a viabilidade (proliferação) de células satélites musculares C2C12 induzidas à diferenciação, mimetizando o processo de reparo após lesão. As células foram cultivadas em Meio Eagle Modificado por Dulbecco (DMEM) suplementado com 10% de Soro Fetal Bovino (SFB) e submetidas à diferenciação pela adição de 2% de soro de cavalo e concomitantemente incubadas com o anabolizante nas concentrações de 5, 10, 25 e 50 µM. Os grupos que não receberam o anabolizante nem o veículo serviram como controle. A viabilidade (proliferação) foi avaliada após um, três e cinco dias, utilizando o método de MTT (3-[4,5-dimetiltriazol-2-il]-2,5 difeniltetrazólio). Context: Bruxism is the habit of clenching or grinding one's teeth in non-functional activities and affects both children and adults alike. Respiratory problems, such as asthma and upper airway infections, are reported to be the etiological factors of bruxism. Aims: The aim of the present study was to determine whether there is an association between respiratory problems and dental caries in children who exhibit the habit of bruxism. Setting and Design: An observational cross-sectional study was carried out. Materials and Methods: Patient histories were taken and clinical exams were performed on 90 children for selection and allocation to one of two groups. For the determination of bruxism, a questionnaire was administered to parents/guardians and an oral clinical exam was performed based on the criteria of the American Academy of Sleep Medicine. Thirty-three male and female children between 4 and 7 years of age participated in the study - 14 children with bruxism and 19 children without bruxism. Statistical Analysis: The data were statistically analyzed using the chi-square test, with level of significance set at 5% (P < 0.05). Results: Mean age of the participants was 5.73 years. The male gender accounted for 45.5% (n = 15) of the sample and the female gender accounted for 54.5% (n = 18). A statistically significant association was found between respiratory problems and dental caries among the children with bruxism. Seventy-seven percent of the children with bruxism had caries and 62.5% the children with respiratory problems exhibited the habit of bruxism. Conclusions: There seems to be an association between bruxism, respiratory problems, and dental caries in children.

ABSTRACT

Background: Chemomechanical removal of carious lesions consists of the dissolution of carious tissue by the application of a natural or synthetic agent, followed by atraumatic mechanical removal. Aim: The aim of the present study was to assess the effectiveness of Papacarie Duo® gel in the chemomechanical removal of carious lesions in primary teeth in comparison to the traditional method (low-speed bur). Settings and Design: A randomized clinical trial was conducted with 20 children between 5 and 8 years of age. Materials and Methods: Two teeth were treated in each child (split-mouth design), with the randomization of two methods: Group 1 – chemomechanical caries removal with Papacarie Duo™; and Group 2 – removal of carious dentin tissue using a low-speed bur. Both methods involved restoration with glass ionomer cement and follow up. The following aspects were evaluated: time required for the procedure; pain (face evaluation scale); rtention of the restorative material in the cavity; and the presence of secondary caries after 30 days. Statistical Analysis Used: Chi-squared test, Student’s t-test, and Wilcoxon test. Results: No statistically significant differences between methods were found regarding time required for the procedure (P = 0.13), the occurrence of pain (P = 0.585), or restoration status at the 30-day clinical evaluation (P = 0.713). Conclusion: The findings of the present study demonstrate that the two methods achieve similar results. The advantages of minimally invasive treatment, such as chemomechanical caries removal with Papacarie Duo™, are its ease of use, patient comfort, and the fact that it causes less damage to dental tissue.

ABSTRACT

CONTEXTUALIZAÇÃO: O músculo esquelético tem a capacidade de adaptação frente a estímulos variados, tais como atividade contrátil, danos diretos e indiretos. Uma das modalidades terapêuticas utilizadas na reabilitação de disfunções musculoesqueléticas que vem demonstrando resultados positivos no tratamento e na prevenção de várias patologias é a terapia aquática. OBJETIVO: Analisar o efeito da natação na expressão dos fatores regulatórios miogênicos MyoD e miogenina durante o reparo do músculo esquelético de rato após criolesão. MÉTODOS: Foram utilizados 40 ratos Wistar, divididos em 04 grupos: (1) Controle; (2) "Sham" (sem lesão, submetido a exposição do músculo tibial anterior (TA); (3) Criolesionado e (4) Criolesionado e submetido à natação, analisados em 7, 14 e 21 dias. A criolesão foi realizada por meio de duas aplicações, utilizando um bastão metálico de extremidade plana, resfriado em nitrogênio líquido diretamente no ventre muscular. O protocolo consistiu de sessões de natação com duração de 90 minutos, realizadas 6 vezes por semana. Ao término do protocolo os animais foram eutanasiados, os músculos TA foram removidos e o RNA total foi extraído. Em seguida, foi obtido o cDNA para a realização do PCR em tempo real utilizando primers específicos para MyoD e miogenina. RESULTADOS: Os resultados evidenciaram uma redução na expressão de miogenina após 7 dias nos grupos criolesionado com (p<0.01) e sem (p<0.01) natação e após 14 no grupo criolesionado com natação (p<0.05) com relação aos grupos controle e "sham", respectivamente. Não encontramos diferenças entre os grupos criolesionados com (p>0.05) e sem natação (p>0.05). Com relação à expressão de MyoD não houve diferença entre os grupos avaliados. CONCLUSÃO: A natação não influenciou a expressão dos fatores regulatórios miogênicos durante o processo de reparo de músculo esquelético de rato após criolesão.


BACKGROUND: Skeletal muscle has the ability to adapt to several stimuli, such as contractile activity as well as direct and indirect damage. Aquatic therapy has been used in the rehabilitation of musculoskeletal disorders. In addition, it has demonstrated positive results in the therapeutic process and preventing several diseases.. OBJECTIVE: The aim of the present study was to analyze the effect of swimming on the expression of the myogenic regulatory factors MyoD and myogenin during the skeletal muscle repair process in rats following cryoinjury. METHODS: Forty Wistar rats were randomly divided into four groups: 1) Control; 2) Sham - non-muscle damaged, submitted to procedure for exposure of the tibialis anterior (TA) muscle; 3) Cryoinjured; and 4) Cryoinjured and submitted to swimming. Analyses were carried out at 7, 14 and 21 days. Cryoinjury was performed with two applications of the flat end of a metal rod previously cooled in liquid nitrogen directly to the belly of the TA muscle. The protocol consisted of 90-minute swimming sessions six times a week. At the end of the protocol, the animals were euthanized and the TA muscles were removed. Total RNA was extracted using the TRIzol reagent. Next, cDNA was obtained to perform real-time PCR using specific primers for MyoD and myogenin. RESULTS: The results showed a reduction in the expression of myogenin in the groups cryoinjury with p<0.01and without p <0.01 swimming after 7 days, and in group cryoinjury with swimming (p<0.05) after 14 days respect to the control groups and "sham", respectively. There were no differences between groups cryoinjury with (p> 0.05) and without (p>0.05) swimming. Regarding the expression of MyoD there was no difference between the groups. CONCLUSION: Swimming did not affect the expression of myogenic regulatory factors during the skeletal muscle repair process in rats following cryoinjury.

ABSTRACT

Aim: To analyze associations between the mouth-breathing pattern and other harmful oral habits among preschoolers. Methods: An observational, cross-sectional study was carried out involving 198 children from 3 to five 5 of age. A questionnaire, clinical evaluation and specific tests (mirror and water tests) were used for confirmation of the mouth-breathing pattern. Results: Mean age of the participants was 4.13 ± 0.8 years and 57.1% were male. A total of 87.4% exhibited one or more harmful oral habits. Harmful habits were more common in the male gender (61.8%); this association was statistically significant (p<0.001). Mouth breathing was the most prevalent habit (49%), followed by biting/sucking on objects (33.3%). Regarding gender, statistically significant associations were found for bottle feeding (p=0.02) and nail biting (p=0.02). Mouth breathing was associated with biting on object (p=0.00), pacifier use (p=0.02) and thumb sucking (p=0.00). Conclusions: The results of the present study suggest that mouth breathing is significantly associated with biting/sucking on objects, pacifier use and thumb sucking in preschoolers and that the occurrence of harmful oral habits is more prevalent among the male gender. Early diagnosis and intervention should be established in order to avoid future consequences involving the orofacial region.

ABSTRACT

Aim: To evaluate the effect to the degree of temporomandibular dysfunctions (TMD) in adolescents and occlusal classes in the cervical posture. Methods: A cross-section, observational study was carried out, in which 296 adolescents took part. For the evaluation, the patients were divided into groups according to the presence and severity of the TMD, using the Helkimo questionnaire and occlusal Angle classification. The posture analysis was carried out using photogrammetry and the software Alcimage® to measure the predefined angle based on the protuberances of the Spinous Process of the 7th cervical vertebra (C7), manubrium of the sternum and mentum vertex. Results: 48% (n= 142) of the 296 adolescents evaluated presented no dysfunction, while 52% (n= 154) presented some degree of TMD. Of the different degrees of TMD, the highest average cervical angulation observed in the group with moderate dysfunction was 97.59º ±7.40, followed by a mild degree, of 96.32º± 9.36, and the lowest average was 93.01º±10.08 in the patients with a severe degree in the different occlusal classes. In relation to occlusal class, higher values for this angulation were observed in class II (96.77º± 8.79), compared with class I (90.64º±8.80) and class III (94.67º± 10.70), a difference which was statistically significant. Conclusions: The subjects with TMD presented a greater alteration in head posture, compared with those without TMD. In particular, the Class II Angle was correlated with TMD and alterations in cervical posture.

ABSTRACT

INTRODUÇÃO: Existe um grande interesse no estabelecimento de recursos e terapias a serem utilizados na tentativa de proporcionar um processo de reparo muscular de melhor qualidade e menor duração. O ultrassom terapêutico (US) e o laser de baixa potência (LBP) são recursos muito usados na prática clínica, porém são escassas, e por vezes contraditórias, as evidências científicas que determinam com segurança os parâmetros dosimétricos e metodológicos adequados. OBJETIVOS: O objetivo do estudo foi analisar o efeito do US e do LBP sobre a proliferação celular durante a diferenciação de mioblastos C2C12. MATERIAIS E MÉTODOS: Os mioblastos foram cultivados em meio de cultura de Eagle modificado por Dulbecco, contendo 10% de soro fetal bovino (SFB), sendo induzida a diferenciação pela adição de 2% de soro de cavalo durante 96 horas. Posteriormente, as células foram irradiadas com US pulsado a 20%, 3 MHz de frequência (intensidades de 0,2 e 0,5 W/cm², durante cinco minutos) ou submetidas ao tratamento com LBP (potência de saída de 10 mW, densidade de energia de 3 e 5 J/cm², por 20 segundos). A proliferação celular foi avaliada após 24h e 72h utilizando o método de MTT. Foram realizados três experimentos independentes, em cada condição citada e células não irradiadas serviram como controle. RESULTADOS: Os resultados obtidos foram submetidos à análise estatística utilizando a Análise de Variância (ANOVA), teste Dunnet, para verificar diferenças entre o grupo controle (não tratado) e os grupos tratados com US e LBP, adotando significância de p < 0,05. Os resultados evidenciaram que não houve diferença significativa na proliferação celular entre as células musculares submetidas a tratamento com ambos os recursos terapêuticos e as células controle, nos períodos de 24h e 72h após tratamento. Além disso, foi possível verificar que não houve aumento significativo no número de células após o período de 72h quando comparado a 24h, confirmando o processo de diferenciação celular, conforme esperado. CONCLUSÕES: Conclui-se que o US e o LBP, nos parâmetros avaliados, não alteraram a proliferação de mioblastos em processo de diferenciação.


INTRODUCTION: There is great interest in establishing resources and therapies to be used in an attempt to provide a process of muscle repair better and shorter. Two features commonly used to facilitate the healing process are the therapeutic ultrasound and laser power, however, are still scarce and sometimes contradictory scientific evidence to determine with certainty the parameters and methodology necessary to acquire these goals. OBJECTIVES: Thus, the objective of present study was to evaluate the effect of therapeutic ultrasound (US) and low level laser GaAlAs 660 nm on the proliferation of C2C12 myoblasts. MATERIALS AND METHODS: The myoblasts were cultivated in culture medium of Eagle modified by Dulbecco, containing 10% fetal bovine serum (FBS) and induced to differentiate by addition of 2% horse serum for 96h. Subsequently, cells were irradiated with US (20%, 3 MHz, and intensities of 0.2 and 0.5 W/cm² for five minutes) or with LLL (output power of 10 mW, density of 3 and 5 J/cm² for 20 seconds). The non-irradiated cells serve as controls. Three independent experiments were performed for each condition cited. Cell proliferation was evaluated after 24h and 72h using the MTT method. RESULTS: The results were statistically analyzed using analysis of variance (ANOVA/Dunnet) to verify differences between control cells (untreated) and US/LLL treated groups. The results showed no significant differences in viability and cell proliferation between LLL or US treated cells and control cells after 24h and 72h. Furthermore, it was observed no increase in the number of cells within the evaluation period after treatment, confirming the process of cell differentiation, as expected. RESULTS: In conclusion, US and the LLL, employed under the parameters described does not alter C2C12 proliferation.

ABSTRACT

El presente estudio se propuso comparar la práctica restaurativa atraumática (PRAT) con el método tradicional (MT) rotacional, con el objetivo de determinar en ambos casos el tiempo total del procedimiento, el costo, la presencia de dolor y el comportamiento en pacientes pediátricos de Perú. De los 30 niños y niñas seleccionados para el estudio, la mitad fueron sometidos a la PRAT y restauración con cemento de ionómero de vidrio y la otra mitad fueron tratados con el MT y restauración con amalgama. Los parámetros de análisis fueron los tiempos necesarios para remover el tejido cariado y el procedimiento operatorio total, el costo total del procedimiento, la manifestación de dolor y el comportamiento del paciente durante el tratamiento. Se hallaron diferencias significativas entre ambas técnicas en todos los parámetros, excepto en el comportamiento del paciente. Si bien la remoción del tejido cariado con el MT fue más rápida, el procedimiento completo fue más rápido en la PRAT, que además resultó significativamente menos costosa y menos dolorosa que el MT. Los resultados indicaron que la PRAT es una muy buena alternativa por su bajo costo y aceptación por parte de los niños.


The purpose of this study is to compare atraumatic restorative treatment (ART) with the conventional rotational restorative method (CM) to determine in both cases the total time required for the procedure, the cost, the presence of pain, and the behavior of pediatric patients in Peru. Of the 30 children selected for the study, half received ART and restoration with glass ionomer cement and the other half, CM and restoration with amalgam. The study parameters were the times required to remove the decayed tissue and to complete the entire procedure, the total cost of the procedure, the presence of pain, and the patient's behavior during treatment. Significant differences were found between the two techniques in all parameters, except for the patient's behavior. Although removing the decayed tissue was faster with the CM, the entire procedure was faster with ART, which, moreover, was significantly less expensive and less painful than the CM. The results indicated that ART is a very good alternative due to its low cost and acceptance by the children.

ABSTRACT

Aim: To analyze gunshot wounds to the face, assessing the characteristics, immediate treatment,late treatment, complications and after effects. Methods: A retrospective observational study wascarried out involving 75 cases of victims of gunshot wounds to the face treated at the Oral andMaxillofacial Traumatology Unit of the Dr. Arthur Ribeiro de Saboya Hospital in the city of SãoPaulo (Brazil).Data analysis used the chi-square test with the level of significance set at 5% (p dd0.05). Results: There was a predominance of the 21-to-30-year-old age group (38.7%), malegender (92%) and wounds occurring due to assaults (37.3%). There was a predominance ofentry wounds on the left side of the face (58.5%). The most affected sites were the mandible(50.7%), maxilla (18.3%), zygomatic region (7.0%), eye socket (4.2%) and nose (1.4%).Comminuted fractures (88.2%) and simple fractures (10.3%) were recorded. No fracture occurredin 1.5% of the cases. The predominant treatment was rigid internal fixation (RIF) (57.2%),followed by exploratory surgery (23.2%) and conservative treatment (19.6%). Among thecases in which the RIF system was used, there was predominance in the mandible (64.0%). Thechi-square test revealed a significant correlation between the 2.4-mm RIF system and the mandiblein 48.0 % of cases. Conclusions: In conclusion, gunshot wounds tended to pierce the face,mainly affecting the mandible and caused comminuted fractures treated with rigid internal fixation.Immediate complications occurred in 25% of cases and after effects occurred in 11.7%.

ABSTRACT

The current study investigated the capacity for tumor factors secreted by head and neck squamous cell carcinoma (HNSCC) cell lines, KB, KB16, and HEP, to induce the secretion of various cytokines from peripheral blood mononuclear cells (PBMCs). PBMCs were isolated from blood samples collected from six healthy volunteers and these cells were incubated for 6, 24, 48, or 72 hours in the presence of 50 percent conditioned medium collected from cultured cell lines pretreated with, or without, stimulants such as phytohemagglutinin (PHA) or lipopolysaccharides (LPS). Aliquots of each supernatant were then assayed for levels of IFN-Γ, vascular endothelial growth factor (VEGF), TNF-α, and IL-4 using enzyme linked immunosorbent assays (ELISAs). Data collected were analyzed using Student's t-test, an ANOVA test followed by Tukey's test, and tests of Pearson's Correlation. PBMCs cultured with KB16-conditioned medium produced the highest levels of IFN-Γ. VEGF was also detected in conditioned media collected from all of the squamous cell carcinoma (SCC) cell lines used, and a significant difference in VEGF levels between control and KB- or KB16-conditioned media was observed. TNF-α was secreted by all PBMC groups within 6 hours of receiving conditioned media, and these levels increased up to the 24 hour timepoint, after which levels of TNF-α stabilized. In contrast, none of the supernatant samples contained detectable levels of IL-4. In combination, these data suggest that direct contact between fresh human PBMCs and conditioned media from tumor cells induces the secretion of TNF-α and VEGF by PBMCs, and this represents an initial angiogenic response.

ABSTRACT

O objetivo do estudo foi investigar a influência da natação sobre as alterações morfológicas do músculo esquelético em processo de reparo após criolesão. Foram usados 45 ratos divididos em cinco grupos: controle (n=5); sham (n=5), adaptação (n=5), criolesionados e tratados com natação sacrificados após 7, 14 e 21 dias (n=15); criolesionados e sem tratamento aquático sacrificados após 7, 14 e 21 dias (n=15). As sessões de natação foram realizadas 6 vezes por semana com 90 min de duração cada. Ao término do protocolo os animais foram sacrificados e a análise morfológica da área da lesão foi realizada. A análise morfológica semiquantitativa demonstrou que os músculos do grupo controle apresentaram aspecto histológico normal. O grupo sham apresentou edema, mionecrose e infiltrado inflamatório em grau 1. Nos grupos 7, 14 e 21 dias, não existiram diferenças estatisticamente significativas nas 4 etapas de remodelamento tecidual avaliadas (infiltrado inflamatório, edema, necrose e fibras musculares imaturas) entre os grupos lesionados quando comparados aos grupos com lesão e tratamento aquático. Em conclusão, foi possível verificar que a natação não causou alterações morfológicas durante o reparo do músculo esquelético após criolesão.


The aim of study was investigate the influence of swimming on the morphological changes in skeletal muscle repair process following cryoinjury. There were used 45 rats divided into 5 groups: control (n=5), sham (n=5), adaptation (n=5), cryolesioned treated with swimming and sacrificed after 7, 14 and 21 days (n=15), untreated and cryolesioned sacrificed after 7, 14, and 21 days (n=15). Animals swan for 90 min/ each session and 6 times a week. At the end of the protocol, the animals were sacrificed and morphological analysis of the lesion area was performed. The semi-quantitative morphological analysis showed that the muscles in the control group exhibited normal histological aspects while the sham group exhibited edema, myonecrosis and inflammatory infiltrate grade 1. In groups 7, 14, and 21 days, the results indicated that there were no statistically significant differences in four stages of tissue remodeling evaluated (inflammatory infiltration, edema, necrosis, and immature muscle fibers) between the injured groups compared to groups with lesion and treated with swimming. In conclusion, it was verified that swimming did not alter morphological aspects of skeletal muscle during the repair process following cryoinjury.

La reproducción correcta de la dimensión vertical de oclusión es la base para la mantención de la salud del sistema estomatognático.Algunos métodos permiten la medición facial de forma reproducible, como la comparación de la distancia Comisura labial – Ángulo del ojo, así como la distancia Nasion-Menton. El objetivo de este trabajo fue evaluar la medida de la Dimensión Vertical de Oclusión y sucorrelación con género en niños y adolescentes. Fueron evaluados 176 individuos, de los cuales 48 fueron selecionados dentro de los criterios de inclusión. La medida de la dimensión vertical de oclusión fue realizada por medio de calibrador digital y fue registrada en fichas individuales. Los datos fueron estratificados por grupo etáreo y analizados por el T-test. Valores de p menores a 0,05 fueronconsiderados significantes. Los resultados evidenciaron que la Dimensión Vertical de Oclusión demostró medidas distintas por grupoetáreo y género, variando de 47,70 mm a 70,20 mm para Ángulo externo del ojo-Comisura labial y 52, 30 mm a 70,60 mm para Nasion – Menton, en el género femenino. Se observó alteración estadísticamente significativa entre estas distancias en las edades de 10 y 12 años en el género masculino, mientras que para el género femenino no fue constatada diferencia significativa, considerando el nivel designificancia de 5%. Se concluye que es necesario el registro correcto de la Dimensión Vertical de Oclusión para la reproducción de esta medida, respetándose la diferenciación por grupo etáreo y género, dado que las alteraciones de la Dimensión Vertical de Oclusión pueden llevar al paciente a alteraciones oclusales, musculares y articulares, además de predisposición a maloclusiones.


The correct reproduction of the vertical dimension of occlusion (VDO) is the basis for restoring health to the stomatognathic system. A number of methods allow facial measurement in a safe, reproducible manner, such as the comparison of distance from the lipcommissure to corner of the eye (LC-CE) to the distance from the base of the nose to the chin (N-Ch). The aim of the present study was to determine VDO length in children between 7 and 12 years of age and the correlation of this length with gender. One hundredseventy-six children were evaluated, among which 48 fulfilled the inclusion criteria. The data were stratified by age and analyzed using the paired Student’s t-test. All p-values less than 0.05 were considered statistically significant. There were different measures of VDOwith regard to age and gender, ranging from 52.30 mm to 70.20 mm for the LC-CE distance and 52.30 mm to 70.60 mm for the N-Ch distance in the girls. There was no statistically significant difference between these distances in the ages of 10 and 12 years for malesand that the correlation between LC-EC and N-Ch was not significant for females. For women, there was a correlation between LC-EC and N-Ch, considering the significance level of 5%. There was conclude that it is necessary to measure the correct VDO and standard measures for reproduction of this measure, while the differentiation by age and gender, since the change in VDO can lead the patientto occlusal changes, muscle and joint pain, beyond the predisposition to malocclusion.

The oral mucous membrane is prone to developing ulcers originating from traumatic or immunological processes. Aim: The aim of the present study was to perform a histological evaluation of the antiinflammatory and healing properties of PapacarieTM applied to oral ulcers. Methods: Fifty adult female albino Wistar rats were randomly separated into two groups: control and PapacarieTM. The animals were anesthetized, placed in prone position and ulcers were induced in the middle dorsum of the tongue through a 3-mm-diameter punch. The control group received no treatment thereafter, while those in the PapacarieTM group received an application of the gel twice a day for 14 days. Five animals from each group were euthanized on days 1, 3, 5, 7 and 14. The tongues were removed, fixed, routinely processed for hematoxylin-eosin staining and analyzed by two blind, calibrated pathologists for the presence or absence of ulcer, inflammatory infiltrate and neutrophils. Data were submitted to statistical analysis by the Fisher’s exact test. Results: On day1, both groups exhibited ulceration and dense acute inflammatory infiltrate. On day 5, reepithelization and scanty chronic inflammatory infiltrate were observed. On day 14, all animals were healed. There were no statistically significant differences between groups with regard to ulceration (p=0.81), inflammation (p=0.55) or neutrophils (p=0.53). Conclusions: Oral ulcers treated with a papain based gel exhibited the same inflammatory reaction and healing aspects as those of the nontreated control group.
